Ingredient Rationale in Depth
Glycine (8g/L)
Glycine is the single most cross-functional addition in the entire formulation:
- Glutathione synthesis: rate-limiting precursor alongside cysteine (from NAC supplementation). Neither alone is sufficient.
- Mast cell stabilization: directly activates the glycine receptor (GlyR) on mast cell surface — a chloride channel that hyperpolarizes the cell, raising the activation threshold for degranulation. This is a completely different mechanism from quercetin (calcium channel) or quercetin (histidine decarboxylase inhibition).
- Collagen: one-third of all collagen by amino acid residue. Functional deficiency in modern diets from elimination of connective tissue foods.
- One-carbon metabolism: substrate for glycine-N-methyltransferase in the SAM cycle.
- NMDA receptor co-agonist: sleep quality improvement (glycine receptor in SCN).
- Bile acid conjugation: improves fat-soluble compound absorption.
At 8g/L × 13ml brine per serving × 3 meals = ~3.1g glycine daily from pickle alone. Still below full therapeutic range (3–10g) but meaningful additive contribution.
Calcium D-glucarate (200mg/L)
Specifically inhibits beta-glucuronidase — the gram-negative bacterial enzyme that deconjugates estrogen glucuronides in the gut, enabling estrogen reabsorption and contributing to estrogen dominance. This connects gut dysbiosis to [[Estrobolome|hormonal dysregulation]] and the autoimmune amplification pathway. Not many interventions address this specific mechanism in food format.
The Emulsification System (lecithin + black seed oil + glycerol)
These three work as a unit:
- Lecithin provides the amphiphilic structure (phospholipid heads face water, tails face oil)
- Black seed oil provides the lipid phase into which lipophilic compounds partition
- Glycerol bridges intermediate-polarity compounds that don't fully dissolve in either
Preparation sequence: warm brine to 40°C → add lecithin granules → add black seed oil → add glycerol → blend vigorously 3–5 min → slight cloudiness confirms emulsion → add salt → add other powders → cool → add to vegetable-packed vessel.
Bacterial Substrates
The molasses provides minor fermentation substrates (glucose, fructose, sucrose). The vegetable matter provides the bulk of fermentation substrate. The Lactobacillus culture does not need supplemental sugar — this is a vegetable ferment, not a sugar ferment.
The Glycine-SAM Connection
The brine is nutritionally active for the Lactobacillus culture as well as for the consumer. Lactobacillus species run their own one-carbon metabolism — the same SAM cycle producing SAH whose recycling produces AI-2 quorum sensing molecules. The glycine in the brine is simultaneously:
- A nutrient for the human consuming the brine
- A potential metabolic substrate for the bacterial culture producing the fermentation compounds
- A component of the one-carbon economy that generates the quorum sensing signals coordinating bacterial community behavior

Glycine and the Bile Conjugation Substrate
The glycine in this brine is doing double duty as a fermentation substrate (the [[Quorum Sensing|AI-2 / SAM connection]]) and as a human bile acid conjugation substrate. A 5ml daily dose of the 3-4x concentrated ferment delivers ~3g glycine from the brine fraction alone, plus additional glycine from the [[Concentration and Harvest|botanical matter]]. The combined dietary + supplement + ferment glycine load of 5-8g daily is the therapeutic range for both glutathione synthesis, mast cell GlyR stabilization, and bile acid conjugation.
