Spice Extraction Methods — Consumer Scale

Which menstruum (extraction solvent) to use, at what ratio, for which compounds, and how to dose the result. The household version of what the [[In-Fryer Antioxidants]] protocol does at the cooking stage.

Why Extract At All

The [[Botanical Pack]] and the [[In-Fryer Antioxidants]] protocol use whole spices in cooking or in the fermentation brine. The in-cook approach is right for sustained low-dose delivery and for fat-phase partitioning during frying. But three situations call for a concentrated extracted form:

  1. Therapeutic dosing — the spice amounts in cooking or fermentation don't reach the doses documented in clinical literature (curcumin 500–1000mg/day, EGCG 400–800mg/day, cinnamaldehyde 100–200mg/day, etc.). A concentrated extract delivers these doses in 1–2 ml volumes rather than tablespoons of spice.
  2. Biofilm disruption work — the [[Upper GI Biofilm Disruption]] protocol requires direct dosing of QS inhibitors and antimicrobials. A tincture is the practical vehicle.
  3. Palatability — concentrated extracts are more tolerable than the equivalent raw material dose.

The Menstruum Decision

For the compounds in the [[In-Fryer Antioxidants]] and [[Botanical Pack]] stack, the relevant logP values determine the best menstruum:

The decision matrix:

  • Ethanol 190-proof (Everclear): best general-purpose menstruum for the polyphenol/terpenoid stack. Captures carnosic acid, thymol, curcumin, eugenol, gingerols, cinnamaldehyde, and berberine at high yield. The 95% concentration is necessary to dissolve the lipophilic compounds; dilute to 70% for water-soluble alkaloid capture if needed.
  • 190-proof vodka (40% ethanol): inferior to 190-proof for the lipophilic stack but safer for skin application and better tolerated by some palates.
  • Apple cider vinegar (raw, with mother): the [[Wild Vinegars]] menstruum. Best for herbal vinegars for daily use. Captures the more polar compounds (EGCG, oleuropein, hydroxytyrosol) and contributes acetic acid + live culture.
  • Extra-virgin olive oil: the high-logP companion to ethanol. Carnosic acid, thymol, curcumin all partition into the oil phase. Adds the food matrix that supports absorption of fat-soluble compounds.
  • Acetone: best industrial solvent for spice oleoresin (paprika, turmeric, vanilla) but the consumer-level problems are real. See below.

The Acetone Question

The vault's [[11-Step Protocol]] uses acetone for fat fractionation — removing PUFA from tallow through selective crystallization. That is a different use case from spice extraction. For spice extraction specifically:

Acetone is technically excellent for the high-logP compounds (carnosic acid, thymol, AKBA, curcumin). Industrial spice oleoresin production uses acetone routinely.

Acetone at consumer level is dangerous because:

  • Vapor is heavier than air, travels along surfaces to ignition sources
  • Stove pilot lights, water heater igniters, static discharge 6+ feet away have ignited acetone vapor
  • Skin/lung irritant
  • Acetone is GRAS for finished food products but the safety protocols for the extraction process are industrial

The vault's own open thread: "Stainless brake line condenser build status unknown" — meaning the recovery still that would make consumer acetone use even remotely safe is not yet built. Until that rig is operational, household acetone extraction is a fire/explosion risk.

Recommendation: use 190-proof ethanol for the lipophilic compounds. Use olive oil infusion for the highest-logP compounds that ethanol under-extracts. Skip acetone at consumer level.

The only exception: if you have built the brake-line condenser recovery still from the [[Acetone Recovery Apparatus]] spec (a future vault note), have electric heat only (no open flames), grounded vessels for static, and adequate ventilation, then acetone becomes a legitimate high-purity option for the high-logP compounds.

The Household Methods

Method 1 — Ethanol Tincture (Primary)

Materials:

  • 190-proof Everclear (or 151-proof vodka for milder result)
  • Dried spices: rosemary, thyme, oregano, sage, cinnamon bark, turmeric, ginger, black peppercorn, clove
  • Glass jar with airtight lid (mason jar works)
  • Coffee filter + cheesecloth for straining
  • Dark glass dropper bottles for storage

Process:

  1. Coarse-grind the dried spices (mortar and pestle or dedicated coffee grinder). The grind increases surface area for extraction.
  2. Combine spices in any proportion matching your therapeutic priority. A balanced "broad spectrum" blend: 3 parts rosemary, 2 parts thyme, 1 part each oregano, sage, cinnamon bark, turmeric, ginger, clove; ½ part black peppercorn.
  3. Pack loosely in glass jar.
  4. Cover with 190-proof Everclear at 1:4 spice:solvent by weight (e.g., 100g spice in 400g ethanol). The 1:4 ratio gives a ~250mg/ml tincture assuming average extraction yield.
  5. Seal, label with date and contents.
  6. Shake daily. Store in dark at room temperature.
  7. Steep 14 days minimum. 28 days is better for the alkaloid and high-logP compound capture.
  8. Strain through coffee filter, then through cheesecloth. Squeeze to recover all liquid.
  9. Transfer to dark glass dropper bottles. Label with date, contents, and ethanol percentage.

Yield: Approximately 80% of the liquid returns. Loss to absorption by the plant material.

Storage: 2+ years in dark glass at room temperature. Ethanol is self-preserving at 70%+ concentration.

Dose: 1–2 ml (about 30–60 drops) with each fat-containing meal. 3 ml is an aggressive dose. Start at 1 ml and titrate up.

The bitterness quality check: The tincture should taste distinctly bitter, complex, and warming. The bitterness IS the analytical confirmation — carnosic acid, thymol, curcumin, gingerols, andrographolide (if present), berberine (if present) are all bitter compounds. A tasteless tincture didn't extract.

Method 2 — Oil Infusion (High-logP Companion)

Materials:

  • Extra-virgin olive oil (or MCT oil for higher saturation)
  • Same dried spice blend
  • Slow cooker or saucepan with thermometer
  • Cheesecloth for straining
  • Dark glass storage bottles

Process:

  1. Combine spices at 1:4 spice:oil by weight in slow cooker or saucepan.
  2. Low heat — 60°C (140°F), 3 hours. Do not exceed 70°C; you'll degrade the more volatile compounds (cinnamaldehyde, gingerols) and risk oxidizing the oil.
  3. Alternative: 14 days at room temperature, shaking daily, with the spices in a cheesecloth pouch for easy removal.
  4. Strain through cheesecloth. Squeeze.
  5. Store in dark glass, refrigerated. Use within 6 months (oil stability limit).

Yield: Approximately 85% of the oil returns.

Dose: 1–2 tsp with each meal. Combine with the tincture for full spectrum — the tincture contributes the more polar compounds, the oil contributes the highest-logP compounds.

Method 3 — Oxymel (Honey + Vinegar)

See [[Wild Vinegars]] for the full method. The honey contributes the [[Honey Microbiota]] climax community + sugar preservation; the vinegar contributes acetic acid + extraction chemistry for the more polar compounds.

Dose: 1 tsp to 1 tbsp in water before meals, or as salad dressing base.

Method 4 — Fresh Allicin Preparation (Special Case)

Allicin cannot be extracted — it forms in situ from alliin via alliinase when fresh garlic is crushed. The protocol is:

  1. Crush or finely mince fresh garlic cloves.
  2. Rest 10 minutes at room temperature. The alliinase enzyme converts alliin → allicin in this window.
  3. Consume with food. The allicin survives stomach acid reasonably well and reaches the upper GI where the QS-inhibitor activity is needed.
  4. Do not cook the crushed garlic — heat destroys allicin. Add to food after cooking, or eat crushed garlic in fermented preparations.

Dose: 1–2 cloves per meal is therapeutic. Higher doses (4–6 cloves daily) are used in biofilm disruption protocols. Can cause GI upset in higher amounts.

The Daily Therapeutic Stack

For a 70kg adult, the combined daily dose from the methods above:

This is approximately the combined dose used in clinical studies on carnosic acid, thymol, curcumin, cinnamaldehyde, and allicin for the relevant indications (mast cell stabilization, QS inhibition, anti-inflammatory, gut barrier).

The Quality Indicators

The taste and trigeminal profile of a correctly extracted preparation:

  • Bitter — quercetin, carnosic acid, thymol, curcumin, berberine, andrographolide. Loss of bitterness = extraction failure.
  • Warming — gingerols/shogaols, piperine, cinnamaldehyde, thymoquinone. TRPV1 activation.
  • Tingle/numb — cinnamaldehyde, eugenol. TRPA1 activation.
  • Pungent bite — allicin (garlic), allyl isothiocyanate (mustard/horseradish/wasabi). TRPA1 + glutathione depletion.
  • Salty-bitter complex — the polyphenol + mineral matrix from the spices.

A preparation that tastes "weak," "flat," or "just sour" is failing. The bitterness and trigeminal activation are the analytical confirmation that the active chemistry is present.

Safety Notes

  • Ethanol interaction with medications: many medications interact with ethanol (metronidazole, certain antibiotics, sedatives). The dose in a tincture (2 ml) is ~1.5g ethanol — small but real. For anyone on contraindicated medications, the oil infusion (Method 2) and oxymel (Method 3) are alcohol-free alternatives.
  • Curcumin and gallstones: high-dose curcumin can mobilize bile and precipitate biliary colic in anyone with existing gallstones.
  • Cinnamon and coumarin: Cassia cinnamon contains coumarin (hepatotoxic at high chronic doses). Ceylon cinnamon ("true cinnamon") has negligible coumarin. For daily therapeutic use, use Ceylon.
  • Allergic reactions: anyone with known Apiaceae family allergies (celery, parsley, carrot, fennel) may react to high-dose herbal extracts.
  • Pregnancy: many of the compounds are contraindicated in pregnancy (especially the berberine, the high-dose rosemary, the androgen-modulating compounds). Lower doses in food preparation are different from therapeutic tincture doses.
